Preparation :

This antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, human cell-derived, recombinant human CD200 ( rh CD200 ; Catalog#10886-H08H ;  NP_005935.4 ; Met 1- Gly 232 ). The IgG fraction of the cell culture supernatant was purified by Protein A affinity chromatography.

Specificity :

Human CD200 / OX-2 

  No cross-reactivity in WB and ELISA with Mouse CD200 / OX-2 and human cell lysate (293 cell line)
Western blot : This antibody can be used at 1 - 2 μg/mL with the appropriate secondary reagents to detect human CD200 in WB. Using a DAB detection system, the detection limit for human CD200 is approximately 4 ng/lane under non-reducing conditions and 8 ng/lane under reducing conditions
Direct ELISA : This antibody can be used at 0.5 - 1 μg/mL with the appropriate secondary reagents to detect human CD200. The detection limit for human CD200 is  0.16 ng/well
Storage : This antibody can be stored at 2℃-8℃ for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20℃ to -70℃. Preservative-Free.
Sodium azide is recommended to avoid contamination (final concentration 0.05%-0.1%). It is toxic to cells and should be disposed of properly. Avoid repeated freeze-thaw cycles.

CD200 / OX-2 Antibody Background

CD200, also known as OX-2, is a transmembrane immunoregulatory protein belonging to the immunoglobulin superfamily. The human CD200 cDNA encodes a 278 aa precursor that includes a 30 aa signal sequence, a 202 aa extracellular domain (ECD), a 27 aa transmembrane segment, and a 19 aa cytoplasmic domain. The ECD is composed of one Ig-like V-type domain and one Ig-like C2-type domain, and shares 76% aa sequence identity with mouse and rat CD200. CD200 interacts with a structurally related receptor (CD200R) expressed mainly on myeloid cells and is involved in regulation of macrophage and mast cell function. CD200 and CD200R associate via their respective N-terminal Ig-like domains. In myeloid cells, CD200R initiates inhibitory signals following receptor-ligand contact. In T cells, CD200 functions as a costimulatory molecule independent of the CD28 pathway. In addition, CD200 also plays an important role in prevention of graft rejection, autoimmune diseases and spontaneous abortion.
